Many cell types require the use of an adherence coating when culturing cells with FRS™ Pioneer—or any serum-free media—on standard TC plasticware. GECKO™ is a chemically defined and animal component free mix of lyophilised adhesion proteins that facilitate attachment of many adherent cell types to standard cell culture plates. GECKO™ supports serum-free adherence of most cell lines traditionally cultured in FBS without a coating. A minority of cell lines will perform optimally with a recombinant vitronectin coating (see below).

Product Information

GECKO™ is a chemically defined, animal component free mix of adhesion factors to support adherent cell culture. Foetal bovine serum (FBS) contains a complex mixture of adhesion-promoting factors that enable attachment on standard tissue culture plastic. When culturing cells in serum-free or chemically defined media, these adhesion cues are often no longer present. As a result, many adherent cell types require an explicit surface strategy to support attachment, spreading, and early survival. GECKO™ has been validated as a defined, animal-free coating which supports a broad range of adherent cell types. Coating-free workflows incorporating GECKO™ have also been validated; reach out to us directly if this is of interest.

GECKO™ was developed to pair with FRS™ Pioneer as part of a fully chemically defined culture workflow.
Using GECKO™ with FRS™ Pioneer allows:

• Separation of growth support (media) from adhesion (surface)
• Greater experimental control
• Reduced background from undefined serum components

GECKO™ can also be used with other serum-free or reduced-serum media systems.

When to use GECKO™
GECKO™ is a good starting point if you:

• Are transitioning from serum-containing media to serum-free or chemically defined media
• Want a single, animal-free coating compatible with many adherent cell lines
• Prefer a reproducible, defined alternative to serum-based pre-coating
• Are culturing adherent cells with FRS™ Pioneer and require attachment support

For cells typically cultured in FBS without a coating, GECKO™ has been heavily validated and is recommended as the first-pass coating. For known exceptions or as a backup, recombinant vitronectin is recommended (see Data & Application Notes).

When to use an alternative coating
You may require a different coating strategy if:

• Your cell line falls into a category previously validated with FRS™ Pioneer for vitronectin coating (see below).
• When culturing your cells in FBS-containing media, you typically use a coating (e.g. collagen, laminin).
• You observe poor attachment, spreading, or survival with GECKO™

In these cases, coatings including but not limited to vitronectin, laminin, fibronectin, collagen, Matrigel or gelatine may be more appropriate.
